Special Techniques using NIS-Elements

Manually separate touching objects

Enclose objects that are not touching

    Remove objects from one channel only

Adjust LUTs for individual objects in a field

Delete Unwanted Objects

Remove NIS if uninstall fails

Measure objects in different channels with consistent numbering

manually separate touching objects...

 

1.                  Open Image and object count window

2.                  Count objects by adjusting threshold, clean, smoothing, and restrictions

3.                  Depress “Keep updating count” button (on far right side of object count window)

4.                  Press TAB key to enter Bin nary editor

5.                  If desired, reduce/increase the transparency of the binary image with “CTRL-down (or up) arrow.

6.                  Select 6-Pt pen line width (near top of screen)

7.                  Toggle “FG” (foreground) to “BG” (Background) in Binary editor

8.                  Select Polyline tool (Left toolbar)

9.                  Left click and hold, draw clear line between touching objects, right click to finish.

10.              Exit Binary Editor

 

To enclose objects that are not touching…

 

Repeat steps 1-7 above

 

7.                  Select Polyline Fill Tool (from left toolbar)

8.                  Left click, draw enclosing circle, right click to finish

9.                  Exit Binary editor, count is automatically updated.

 

Remove Objects from one channel of a Multi-Channel image...

This technique was successfully used to remove a  large diffuse red blotch from a FISH cell while retaining the distinct red dots.

REMOVE RED OBJECTS FROM WITHIN INDIVIDUAL OBJECTS

1.                  extract red channel top work on

2.                  reference…current image>reference (to activate image Arithmetics)

3.                  Binary...Binary editor (or tab key)

4.                  Polygon fill tool (F4) Circle object to keep

5.                  Exit Binary editor

6.                  Reference…Image Arithmetics…Current AND Binary

7.                  Uncheck “View Overlay” Button

 

RETURN TO ORIGINAL IMAGE

8.                  Remove red channel

9.                  drag modified red channel back onto original image

 

Adjust LUTs for individual objects in a field...

    1.        Open Fresh Image

 2.      reference…current image>reference (to activate image Arithmetics)

3.      Binary...Binary editor (or tab key)

4.      Polygon fill tool (F4) Circle object to keep

5.       Exit Binary editor

6.      Reference…Image Arithmetics…Current AND Binary

  1. Uncheck “View Overlay” Button
  2. Adjust LUT and Burn in change
  3. Reference…Image Arithmetics…Current+Reference

 

Delete unwanted objects such as dirt and debris from around other objects in brightfield...

     1.   Fresh Image

2.      reference…current image>reference (to activate image Arithmetics)

3.      Binary...Binary editor (or tab key)

4.      Polygon fill tool (F4) Circle object to keep

5.   Exit Binary editor

6.      Reference…Image Arithmetics…Current AND Binary

  1. Uncheck “View Overlay” Button

To Remove NIS-Elements when the Uninstall shield doesn't work...

1.    Delete the NIS-Elements Folder

2.    Run the Clean_Util.zip program

3.    Delete the following folder... 

C:\Program Files\InstallShield Installation Information\{CD229881-9012-4B72-9FF7-6719C44B5CB0}

(InstallSheild Installation is a hidden folder so make sure you have the option to see hidden folders on).
